Project Goals

Based upon traditional research methods, we believe that there are at least three large families of Whitneys in the United States along with potentially many other much smaller groupings. These three main branches, descendants of immigrant John Whitney of Watertown, descendants of Henry Whitney of Long Island, and descendants of Samuel Whitney of Bermuda may or may not have been related in the generations before they came to the new world.

Goal 1 - Determine standard "signature" DNA profiles for John, Henry, and Samuel - COMPLETE

This will tell us if these groupings are related or not, and once we have them, will allow us to inform unconnected individuals who are tested which group they belong to.

Goal 2 - Identify where DNA changes occurred to assist "unconnected" Whitneys (people descended from ancestors with the Whitney surname who cannot link to one of the known family lines) in determining where they probably connect.

Goal 3 - Find descendants with known origins in specific England localities to help in determining where the immigrants originated from

The DNA project will try to establish benchmark Y-DNA profiles of each of the four lines. If the lines are related, we will find that the Y-DNA is the same for all of the related lines. If the Y-DNA is off by one marker, there may well have been a mutation back in the past, which may enable us to differentiate the different lines, even if they are related. If the Y-DNA is different by more than 2 markers, the lines are not related (or at least not closely enough to matter).

How does DNA Testing Work?

First, a quick overview of how all of this DNA stuff works. Each time a child is conceived, he or she inherits an X chromosome from the mother, and either an X or a Y chromosome from the father. If the child receives an X from the father, then it has two X chromosomes (XX) and develops into a daughter, but if the child gets a Y from the father (XY), it develops as a son. This is the reason that the Y chromosome is used for DNA studies - it comes from the father to the son with no mixing from the mother.

Now think of the Y chromosome as a huge computer program - a program that is turned on and off at various times in the lifetime of the individual, and on and off at different places in the body (the program code to build liver cells does not need to be activated in the foot, for example). During development, the Y chromosome is what causes the changes which result in a boy instead of a girl. During any generation, there could be a random copying mistake made in the DNA. If this mistake causes a problem in a critical area, the child may have a disease or birth defect, or if very severe, may be stillborn.

Now in this large computer program, there are 'junk' areas which don't really do anything useful. If a change happens here, there is no harm done to the baby. Scientists have found such areas where a pattern seems to repeat over and over. Imagine something like this: "EndOfLastProgramCodeDubbaDubbaDubbaDubbaStartOfNextCode". See the "Dubba" repeat there? There are 4 repeats of "Dubba", and so if this spot was given a label, the value would be listed as "4".

There are now 67 places where scientists have found repeats like this, and they have been given names like "DYS 393", and they have found that people can have 12, 13, 14, etc. repeats at this spot. On rare occasions, when a father transmits his genetic information on to a child, the number of repeats can be copied incorrectly, so a father may have 11 repeats while the son only has 10 repeats.

By analyzing a number of descendants of John Whitney, for example, we have been able to see what the most common value is at each of the 67 markers and designate this as the immigrant John Whitney's "signature" profile - the values at these markers that we believe that he had. By comparing the random changes in these values with known lines of descent, we can estimate where those changes likely occurred. Once we know this, we can assist people with unconnected Whitney lineages in determining where they should concentrate their research.

How do I Participate?

We have selected FamilyTreeDNA to act as our lab for the testing. They offer 12-marker, 25-marker, 37-marker, and even 67-marker DNA testing. The cost for the 12-marker test is $99, a group rate available to projects like this one. Since they retain your DNA samples at their lab, it is possible to upgrade to a higher level test if you decide to learn more. We will be starting with the 12-marker test first in the interest of economy and because it will show if there is the likelihood of a common ancestor. If you participate, you will be able to sign up directly for the project and pay for the testing kit by credit card (even if you live outside the US).

To do the testing, first we will need contributions of DNA from males who are in direct line back as far as we have good records. I would like at least 3 from each line, as distant from each other as possible but within the line. Once we have established a benchmark Y-DNA profile for the line, it will be possible for any male, connected or not, who thinks he is in the direct line from any Whitney ancestor, to determine if he is, and if the lines are not all related, to which line. If all of the lines are related to each other, however, the best this test will do is to indicate that he is a part of the family. It cannot differentiate unless there is either a mutation or an unrelated line. Unfortunately, this does not work for the women. Y-DNA is only transmitted father to son.

If you choose to allow direct contact between you and other members of the project, you must enclose a signed release form with your submitted sample. Otherwise, all contact with others will be indirect, through me as Group Administrator.

Cladogram

Another way of viewing the information is to create a Cladogram. These charts determine an interconnection of all the DNA signatures using the minimal number of mutations (rule of parsimony). It may or may not accurately depict ancestor-descendant relationships that a genuine tree would show, but it is useful, however, for evaluating potential relationships.

This chart was created using a free phylogenetic network software program offered by Fluxus Engineering http://www.fluxus-engineering.com/sharenet.htm. This program determines the simplest configuration which has the least number of interconnections or mutations. In the chart shown below, the large yellow circle in the center a number of descendants of the immigrant John Whitney who share his DNA signature. Radiating out from the central circle are spokes which connect to smaller circles. These circles are men whose haplotype is different from John's haplotype by one or more mutations. The black number beside each circle represents FTDNA kit number. Some of these have been replaced with a name, such as "MRTNSY" for the line from Morton Say, England. This style of charting nicely identifies the "modal" (meaning most common) haplotype and the mutations which radiate away from that haplotype.
